ρ-COUMARIC ACID

A NATURAL ANTIOXIDANT and antimicrobial agent;

ρ-Coumaric Acid is a naturally occurring polyphenol found in wine, vinegar, honey, tomatoes, basil, garlic and fruits.

ρ-Coumaric Acid is a potent antioxidant that can inhibit lipid peroxidation and radical scavenging as well as an effective antimicrobial agent against E. coliand S. aureus. When applied in cosmetic products, it also has skin lightening effect.

The ρ-Coumaric Acid was tested for antibacterial potential against Gram-positive (S. pneumoniae ATCC49619,B. subtilis 9372,S. aureus 6538) and Gram-negative bacteria (S. dysenteriae 51302,E. coilATCC25922, Salmonella typhimurium 50013). The MIC values for ρ-Coumaric Acid are shown in the table above. A comparison of the sensitivities of the bacterial strains toρ-coumaric acid indicated that the strongest inhibitory effect was againstS. dysentenae. ρ-Coumaric Acid showed MIC value of 10μg/ml in the case ofS. dysenteniae.

ρ-Coumaric Acid has the optimal structure to be a compettive inhibitor of tyrosinase that catalyzes key reactions in the melanin biosynthetic pathway, thus it has good potential to be used as a skin-lightening active ingredient in cosmetics.

ρ-Coumaric acid can attenuate skin hyperpigmentation through multiple mechanisms. UV and other pathophysiological conditions stimulate the production of reactive oxygen species (ROS) and multiple signaling pathways leading to enhanced gene expression of tyrosinase and increased melanin synthesis. The melanin can absorb ultraviolet (UV) radiation and alleviate oxidative stress and inflammatory reactions caused by UV radiation, but the melanin deposition may cause skin pigmentation disorders.ρ-Coumaric acid has a chemical structure similar to L-tyrosine and inhibits the activity of tyrosinase, which catalyzes the oxidation of L-tyrosine and/or L-DOPA to L-DOPA quinone in the melanin biosynthetic pathway. Due to its UV absorption and antioxidant action,ρ-coumanic acid can inhibit the signaling pathways linked to gene expression of tyrosinase and infiammatory mediators.ρ-Coumaric acid can also reduce the stimulatory effects of hormones and L-tyrosine on the gene expression of tyrosinase. Thus, it is proposed thatρ-coumaric acid has advantageous biochemical properties suitable for use as a skin-lightening active ingredient in cosmetics.
